Linkage– Tandem MS fragmentation can be used to quickly determine linkage 2,3 vs 2,6 linked sialic acid based on diagnostic fragmentation ions typical to each linkage. Critical data regarding the sialylation of an engineered or isolated glycoforms is obtained. Alternatively, enzymatic method for determining 2,3 vs 2,6 linked sialic acid may be used in conjunction with HPAEC-PAD monosaccharide analysis.
